Could someone help to understand how to add transport cost to cart?
Is it possible to choose from different transporters cost like some online shops support?

I found that it can be done via Tax Rules.
My question now: Is it possible to add Tax and then Freight cost of R350.00 if Net amount < R1000.00?

It is done through Shipping Rules. Tax Rules are for taxes, not for shipping. Bear in mind that there can only be one shipping rule for the whole Country. You can’t setup shipping cost as per States, Cities or Postal codes. I have asked a similar question here.
