How do I generate e-Way Bills from ERPNext?
After going through some references online (like this one: Eway Bill - What is Eway Bill? E Way Bill System, Rules and Generation Process Explained) an E-Way Bill can only be generated from the Government Portal or SMS (registered from said portal).
In ERPNext you can generate a Material Transfer Receipt to go along with the e-way bill.

Is there any possiblity to genrate the e-Way bill JSON for purchase invoice?
Have you tried the API link of purchase invoice? It should be easy to even make a custom method to be accessible from the web.
