Yes, items can be present in multiple warehouses. You can create a Stock entry and specify and item and the warehouse.

What would be the stock entry type??
You can choose Material Receipt. This means you are receiving stock in your warehouse. The source warehouse in this case is not mandatory. If you wish to transfer stock from one warehouse to another, choose the source and target warehouse.

And how can we select those two warehouses by default in pick list of that item?
