I was wondering if Report builder could support the below.
It is like GUI based Query report.

ERPNext allows different ways to create new and customize existing reports. Something similar to a GUI based approach to customize a report is available using “Report Builder”.

No we cannot but I’m not sure what you mean by grouping columns. Since a column represents a certain field or value for certain doctype (like Net Total for Purchase Invoice or Gross Profit for Sales Invoice)
